Prerequisites: Attendance of the microbiology course
Target skills and knowledge: Basic notions of gene therapy approaches; knowledge of the main viral vectors used in gene therapy; current applied technologies; perspective of gene therapy approaches design.
Examination methods: Oral
Assessment criteria: Knowledge of the subject treated and ability to argue and justify the use of various systems for gene therapy; scientific language and terminology skills.
Course unit contents: -Introduction to gene therapy: choice of therapeutic gene and method of administration of the therapeutic gene.
- Nucleic acids with therapeutic function: genes encoding proteins and non-coding nucleic acids.
- Methods of gene transfer: physical methods and chemical methods; viral vectors for gene transfer: gammaretrovirus-based vectors, lentivirus-based vectors, vectors based on adeno-associated virus, adenovirus-based vectors, vectors based on the herpes simplex virus.
- Examples of clinical trials using viral vectors.
- Tumor gene therapy: active, passive and adoptive immunotherapy; gene therapy with suicide genes; use of oncolytic viruses.
- "Gene editing" techniques for gene therapy: introduction to "gene editing", potentiality and use in clinical trials, use of artificial nucleases, Zinc Finger nuclease, Transcription activator-like effector nucleases and CRISPR / Cas9 system.
Planned learning activities and teaching methods: Lectures and studies of recent scientific publications in the field of gene therapy.
