Syllabus
Prerequisites: Students must know the structure of DNA, RNA and proteins, the concept of the genome and the fundamentals of biochemistry
Target skills and knowledge: The aim of the course is to show recent techniques of purification and characterization of proteins and peptides. This course is designed to let students explore some of the many biochemical methods that are utilized for elucidating structural and functional properties of proteins. The practical laboratory introduces and emphasizes important techniques in biochemistry, including, 2D-electrophoresis, liquid chromatography and mass spectrometry. A large part of the course is dedicated to provide knowledge of proteomics and proteomics techniques.
Examination methods: Final oral exam and evaluation of the results obtained during the laboratory. A recent article is also discussed.
Assessment criteria: The aim of the exam is to verify the knowledge of aspects relating to the analysis of a proteome. The knowledge of modern mass spectrometry instruments will be also verified.
Course unit contents: The course covers topics that concern both the chemical characterization of proteins and the study of a proteome. In particular are presented techniques ranging from the analysis of a protein fingerprinting and sequence determination by Edman degradation, to the separation of complex mixtures of proteins by 2D-electrophoresis and multi-dimensional chromatography. A large part of the course is dedicated to the mass spectrometry of proteins. The main techniques of mass spectrometry and mass-mass of proteins are presented, both from the point of view of the theoretical aspects from that of interpretation of mass spectra.
Another topic addressed is bioinformatics in proteomics. As specific applications of mass spectrometry will face the determinations of post-translational modifications. Finally, topics such as analysis of chemical modifications of proteins, Quantitative Proteomics,Functional Proteomics are also treated.

The laboratory training is performed in equipped laboratories and each student has available personal equipment, to carry out his work.
