Prerequisites: Organic Chemistry 1, Organic Chemistry 2.
Target skills and knowledge: The course completes the preparation in basic organic chemistry initiated with the preceding two courses. In addition it introduces NMR and mass spectrometry for the identification and structural characterization of organic compounds.
Examination methods: Written exam constituted by two tests with multiple-choice or open questions, a simple retrosynthesis/synthesis and, for the first part, the solution of a simple problem using given spectral information. The final grade will be the result of the average of the two test to which the lab score will be added.
Assessment criteria: Knowledge of the course subjects: level of correctness of the answers; correctness of problem solutions. Laboratory proficiency.
Course unit contents: Part 1: physical methods in organic chemistry.
Nuclear Magnetic Resonance (NMR): introduction of principles and procedures. Proton NMR: chemical equivalence and chemical shift; scalar coupling; spin systems; magnetic equivalence; analysis and interpretation of first order proton NMR spectra; decoupling. Carbon-13 NMR: analysis of broad-band proton decoupled NMR-C-13 spectra.
Mass spectrometry: introduction of principles and instrumentation for electron ionization experiments; isotopes; fragmentation; molecular ion and base peak; characteristic fragmentation reactions of major classes of organic compounds. Exercises of spectra analysis and interpretation.
Part 2: organic chemistry.
Acid-base chemistry of carbonyl compounds; inductive and resonance effects; keto-enol tautomerism; enols and enolates; alfa-halogenation of carbonyl compounds and the haloformic reaction; enolates nucleophilic addition to carbonyl compounds and condensation reactions (aldol reactions, Claisen reaction); synthesis of alfa,beta-unsatured carbonyl compounds; 1,2 and 1,4 nucleophilic addition; Michael addition; Robinson annulation; decarboxylation. Reduction and oxidation reactions: hydrogenations, reduction with metal, reductions with hydrides; oxidations with metals, with catalysts, with peroxides and hydrogen peroxide. Stereoselectivity in the oxidation reactions. Introduction to heterocyclic compounds. Pericyclic reactions: cycloadditions, electrocyclic reactions, sigmatropic rearrangements
Planned learning activities and teaching methods: Lectures in class (theory and exercises) with the aid of slides projection.
Laboratory experiment: characterization of unknown compound through chromatographic separation and acquisition and analysis of proton NMR and mass spectra.
