Target skills and knowledge: The course is organized in lectures and practical exercises in the laboratory. It aims at to providing the basic knowledge for the qualitative and quantitative analysis of the active ingredients of medicinal and aromatic plants with particular regard to their derivatives. Students will learn the main analytical methods for the recognition of the active principles of plants with traditional chemical methods and instrumental techniques such as UV-Vis spectrophotometry, fluorimetry, Infrared, Polarimetry, Gas Chromatography, HPLC, Atomic Absorption and Mass Spectrometry
Examination methods: Lectures and laboratory exercises where the student is required to complete a report on work carried out by evaluating the data he obtained as compared with those actually delivered
Assessment criteria: Written examination with assessment of laboratory experiments and subsequent discussion.
Course unit contents: Methods for chemical analysis of the major classes of phytochemicals. Theoretical basis of the general methodologies for the qualitative determination of the various classes of phytochemicals, precipitation reagents, staining reagents, methods of separation of substances in phytochemicals, Theoretical basis for the quantitative determination of the substances present. Gravimetric methods, titration methods: acid-base, redox, complexometric, precipitation. Evaluation of experimental errors,calibration curve method and multiple addictions method.

Determination flavonoids components, determination of fat acidity, of peroxide value, saponification and esterification indees. Characterization and dosage of xanthine bases, study of phytochemicals used as colorants, dosage of amino acids and vitamins.
Planned learning activities and teaching methods: Introduction to the qualitative analysis of the main functional groups present in phytochemicals.
Phytochemical screening, systematic approach to qualitative analysis, extraction, isolation of compounds. Introduction to the methods of quantitative analysis, systematic errors, random errors, mean median, standard deviation. Material and calibrated equipment, methods of gravimetric analysis, mineralization of compounds and determination of the amount of isolated compound. Volumetric titration methods: acid-base titration, redox, complexometric, precipitation, according to the indicators, the equivalence point and end point, use of the melting point. Classification of the main classes of phytochemicals and identification essays; alkaloids, glycosides and carbohydrates, cardioactive glycosides, flavonoids, phenols and alcohols, esters, tannins, esters of fatty acids, anthraquinones, anthocyanins, xanthine bases, saponins, essences and essential oils. Introduction to optical methods of instrumental analysis: UV-Vis, fluorimetry, Atomic Absorption, Polarimetry, Infrared, NMR qualitative and quantitative analysis. Mass Spectrometry and chromatographic separation methods Gas-Chromatography, HPLC. In the practical part of tlaboratory activities are provided individual and group. Introductory lessons on safety in the laboratory. A series of experiments relating to the identification and quantification of some substances.
Identification of sugars (chemical assays, melting point, polarimetry, IR), xanthine bases (UV-Vis, HPLC), flavonoids saffron, caffeine (chemical assays, UV-Vis, HPLC) and catechins in black and green tea, identification and determination of citric acid and ascorbic acid in blackcurrants, acidity dosage of seed oil, dosage of amino acids with ninhydrin method, determination of copper by atomic absorption, identification and determination of essential oils with polametry, IR, and GC mass.
