Syllabus
Prerequisites: Knowledge on cell biology, pathology and microbiology are needed.
Target skills and knowledge: The main biotechnological techniques and the most important classes of biopharmaceuticals approved in therapy or in advanced clinical trials, will be treated.
Examination methods: An oral examination will be taken to demonstrate the knowledge of both the biotechnological techniques and biopharmaceuticals.
Assessment criteria: The objective is to test the student’s ability to understand the principles of the pharmaceutical biotechnologies and to knowledge the biopharmaceuticals with particular attention to the differences with respect to conventional drugs.
Course unit contents: The content of the course is divided in two parts: in the first one the most used biothecnologies will be discussed, in the second part the biopharmaceuticals or the classes of biopharmaceuticals most important from a therapeutic point of view will be described.
The topics of the first part will be:
- recombinant DNA technology: vectors, restriction enzymes, transformation, insertional inactivation. Examples of drugs prepared by recombinant DNA technology.
- DNA sequencing: chemical and enzymatic methods.
- Polymerase chain reaction.
- Nucleic acid technologies: antisense, triple helix, aptamers and ribozymes. Examples of drugs prepared by nucleic acid technologies.
- Hybridoma technology for antibody production. Chimeric, humanized and human monoclonal antibodies: phage display and knockout mouse.
- Glycotechnology: native and modified erythropoietin, heparins.
- Gene therapy. Viral and non-viral vectors. Retrovirus, adenovirus and adenovirus-associated. Diseases targeted by gene therapy and clinical trials.
In the second part of the course the following biopharmaceuticals will be described:
- cytokines: interleukins and interferons.
- Hormons: native and genetically engineered insulin, growth hormon.
- Monoclonal antibodies in anticancer, immunosuppressive and antiviral therapy.
- Enzymes: native and recombinat tissue plasminogen activator, human recombinant DNAse.
Planned learning activities and teaching methods: Frontal lecturing with audiovisual supports (mainly slides) available on MOODLE platform.
